Design Notes

Since we are using the U6 promoter to create our untranscribed mRNA strands, there has to be a Guanine present in front of our target sequence.

The U6_target sequence plasmids were synthesized by IDT and blunt end cloned into Invitrogen's pCR-BluntII-TOPO plasmid. Our U6_target sequence follows the U6_guide RNA design published by the Harvard Church Lab.
